Toxicology of opioid analgesics and their impact on traffic accidents in the Moravian-Silesian Region
DOBIÁŠOVÁ, Marcela
The bachelor thesis is devoted to the issue of the use of opioid analgesics, their effect on traffic accidents and accidents caused under the influence. Opioid analgesics, which belong to the group of narcotic and psychotropic substances, are still a topical topic, as a large number of traffic accidents are caused by drivers who are under their influence. The theoretical part is devoted mainly to history, classification of opioid analgesics, distribution of narcotic and psychotropic substances, list of drugs containing narcotic and psychotropic substances in the Czech Republic, occurrence of side effects, addictions, route of administration, detection of addictive substances in the body, but also drug policy strategy in Moravian-Silesian Region, the Czech Republic and the European Union. The practical part is focused on the comparison of traffic accidents caused under the influence of opioid analgesics in the years 2014-2018 in the Moravian-Silesian Region. In the practical part, I processed the results based on the statistics of the Police of the Czech Republic. After evaluating the results, it was found that in the period 2014-2018, the number of traffic accidents under the influence of opioid analgesics increased in the Moravian-Silesian Region, but there was a decrease in traffic accidents caused under the influence of alcohol. At the same time, there was a reduction in the number of people who were slightly injured, severely injured and killed. The result also showed that the most accidents that were caused in the period under the influence of addictive substances were in the South Moravian Region.
Regulation of advertisements in pharmaceutical industry
Metelka, Jan ; Patěk, Daniel (advisor) ; Rozehnal, Aleš (referee)
Abstract, Jan Metelka The main aim of this diploma thesis was to firstly explain the field of human pharmaceuticals and regulation of advertisement regarding them, from the general point of view and then in details. In order to achieve this it was required to define advertisement and pharmaceuticals because these terms and crucial for the whole thesis. Both definitions may be off the scope of this text however it plays an important role in understanding all necessary issues. Advertisements in pharmaceutical industry have some specifics as described in chapter 2. There are two kinds of recipients - experts (doctors and pharmacists) and ordinary people (mostly patients). Every kind is special and regulated in a different way. Third chapter described all steps in administrative proceedings related to Act on Advertisement, forming a public branch of law against unfair competition. It summarises and divides all misdemeanours, statistically describes how often they occur and deeply analyses possible breaches of Act on Advertisement. The main part of this thesis is Chapter seven, dealing with unfair competition in private law, as mentioned in early chapters, however here it is specifically described how to apply the general clause and the more important kinds of unfair competition such as deceptive adverts and...
Determination of Lipophilicity of Drugs by HPLC
Pleváková, Magdaléna ; Kubíček, Vladimír (advisor) ; Nováková, Veronika (referee)
3 ABSTRACT Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of biophysics and physical chemistry Author: Magdaléna Pleváková Supervisor: Ing. Vladimír Kubíček, CSc. Thesis title: Determination of Lipophilicity of Drugs by HPLC In this thesis a RP-HPLC method for fast and reliable determination of lipophilicity was proposed and tested. Stationary phase was selected by using hydrophobic substraction model. Capacity factors of the chosen substances were initially measured on Zorbax ECLIPSE XDB C18 250x4,6 mm, 5µm column, which exhibits almost identical retention characteristics as the column used for this purpose until now. Then the capacity factors of the same substances were determined by using Zorbax ECLIPSE XDB-C18 50x4,6 mm, 1,8µm column that was selected to reduce retention times significantly. A group of newly synthesised drugs based on structure of pyrazine served as samples for the measurement. The reproducibility of the capacity factor values determined using both columns was compared and the independence of the capacity factor on the mobile phase flow was confirmed. The capacity factors of two homologous series and a group of benzimidazols were consequently determined on Zorbax ECLIPSE XDB-C18 50x4,6 mm, 1,8µm column using various compositions of mobile phases. Several...

The diploma thesis deals with witchcraft and other mystic phenomena in conception of Edward Evan Evans-Pritchard. It is focused especially on the notion of witchcraft in connection with magic, shamanism and oracles. It describes relation of all these phenomena towards religious belief and various attitudes towards them. It defines respective practices and explains differences between them. Last but not least, it introduces usual reactions to them. Via concrete situation it also shows interconnection of all described phenomena and presents them as a coherent and logical system.
Physico-Chemical Properties of Drugs
Tomanová, Pavla ; Kubíček, Vladimír (advisor) ; Nováková, Veronika (referee)
Charles University Faculty of Pharmacy in Hradec Králové Department of biophysics and physical chemistry Author: Pavla Tomanová Supervisor: Ing. Vladimír Kubíček, CSc. Thesis title: Physico-Chemical properties of drugs This thesis deals with measuring dissociation constants (the pKa value) and the lipophilicity of nitrogenous heterocycle derivatives produced at the Faculty of Pharmacy in Hradec Králové. Two methods were used to determine the pK value: potentiometric titration and spectrophotometry. Potentiometric titration almost invariably produced relatively exact results, however the results of pK value obtained by spectrophotometry were sometimes difficult to replicate. Dissociation constants were compared on the basis of the method applied, and also with the theoretically determined dissociation constant calculated by means of the ChemDraw Ultra 12.0 software. The results obtained by the two experimental methods turned out to be very similar, corresponding to the theoretical pK value. Lipophilicity is another essential property of drugs tested in the preclinical stage. It specifies the aqueous or lipophilic phase affinity of a drug, substantially affecting the drug's ADMET in organism. Lipophilicity was determined on the basis of capacity factor value identified by RP-HPLC measurement using...
Multicomponent Analysis
Kovářová, Lucie ; Kubíček, Vladimír (advisor) ; Drastík, Martin (referee)
Charles University in Prague, Faculty of Pharmacy in Hradec Králové Department of Biophysics and Physical Chemistry Candidate: Lucie Kovářová Supervisor: Ing. Vladimír Kubíček, CSc. Title of Diploma Thesis: Multicomponent analysis This diploma thesis deals with the determination of concentrations of individual dyes in mixtures by multicomponent spectrophotometric analysis. The method was tested on selected dyes which were divided into two groups. The first group contains methyl orange, methyl red, orange II and phenol red. The second group consists of methylene blue, bromocresol green and thymol blue. Within each group mixtures were prepared containing combinations of two, three and four dyes in various ratio. Spectra of prepared mixtures were measured by diode array UV/VIS spectrophotometer. Absorbance values were read at wavelengths of absorption maxima of the individual dyes. Using the pseudoinverse matrix, concentration ratios of components were calculated from the absorbance values. The determination in determined and over determined systems was performed. On the basis of the results obtained by the method of analysis it is possible to determine the individual concentration of the multi-component dyes mixtures without prior separation. Keywords: spectrophotometry, multicomponent analysis,...
Cyclodextrin-drug conjugates equipped with targeting groups as anticancer agents
Lamačová, Lucie Josefa ; Jindřich, Jindřich (advisor) ; Smrček, Stanislav (referee)
This diploma thesis deals with the synthesis of conjugates of cyclodextrin with the anticancer drug 5-fluorouracil and folic acid, which works as a targeting group. 5-Fluorouracil is connected to cyclodextrin via an acid-labile linker, which is expected to be cleaved in decreased pH in the proximity of malignant tissue or in the endosome. Malignant tissue also overexpresses receptor for folic acid, and this phenomenon is used for targeted delivery of therapeutic agents. Cyclodextrins are cyclic oligosaccharides, which are known for their ability to complex various compounds into their hydrophobic cavity and increase solubility, stability and bioavailability of these compounds. A synthetic approach for the preparation of conjugate of cyclodextrin with 5-fluorouracil and folic acid was designed and the conjugate was subsequently synthesized. Key words: cyclodextrin, fluorouracil, targeting group, folic acid, drug delivery
Biodegradable polymer carriers with structure tailored for increased tumor tissue permeability
Hrochová, Michaela ; Etrych, Tomáš (advisor) ; Sedláček, Ondřej (referee)
Cancer diseases are the most common cause of death over the world. One fifth of death is caused by cancer in the Czech Republic. The big problem in the treatment of cancer diseases are side effects of chemotherapy, which make treatment more difficult for patients. The present bachelor thesis is focused on synthesis and characterization of novel polymer carriers. Polymer carriers can target the cytostatic drug to the tumor tissue and minimize side-effect of chemotherapies. This bachelor thesis is focused on synthesis and physical chemistry characterization of new diblock polymer carriers, specifically carries of docetaxel. Successful synthesis of novel water-soluble polymer precursors and conjugates based on N-(2-hydroxypropyl)- methacrylamide is described and discussed in this thesis. The hydrolytic stability of the synthesized diblock systems was studied in 0,3M phosphate buffer at pH 7,4 simulating the bloodstream environment. It has been successfully verified that the synthesized polymer systems are hydrolytically degradable after fulfilling its role of drug carrier and therefore can be finally excreted from the organism. The effect of the type of oxoacid used for modification of the cytostatic on the release rate of the carried cytostatic from the polymeric carriers was thoroughly evaluated and...
